A sample of intact mitochondria is incubated in a buffered solution containing succinate (an oxidizable substrate) and ADP plus P$_i$. Oxygen consumption and ATP production are monitored. Upon the addition of cyanide (an inhibitor of Complex IV, cytochrome oxidase), scientists re-measure the oxygen consumption and ATP productions. The results are shown in the graph below.
